MetaVia Announces Completion of Dose Titration in Phase 1 Part 3 Study of DA-1726 for the Treatment of Obesity

MetaVia Inc., a clinical-stage biotechnology company focused on addressing cardiometabolic diseases, has announced that all active patients in Part 3 of its Phase 1 clinical trial evaluating DA-1726 for obesity treatment have successfully reached the highest target doses of 48 mg and 64 mg. DA-1726 is an oxyntomodulin analog designed to target GLP-1 and glucagon receptors. The trial includes two 16-week cohorts to study one-step and two-step dose-escalation strategies to improve tolerability while reaching higher doses.

Hyung Heon Kim, President and Chief Executive Officer of MetaVia, commented on the milestone, stating that the completion of dose titration in both cohorts reinforces the drug's favorable tolerability profile. He expressed confidence that the company's titration strategy could provide a notable competitive edge over existing obesity treatments.

The trial aims to evaluate safety, tolerability, pharmacokinetics, and pharmacodynamics of DA-1726. The planned enrollment is approximately 40 adults, divided into two cohorts with a 4:1 randomization of active versus placebo. Part 3A involves a one-step titration regimen, while Part 3B uses a two-step titration regimen. Primary endpoints will include monitoring various types of adverse events, and secondary endpoints will assess metabolic and other cardio-metabolic measures, such as weight and waist circumference.

The topline data for the trial is expected to be reported in the fourth quarter of 2026. In previous studies, DA-1726 demonstrated potential benefits in weight loss and metabolic improvements compared to leading GLP-1 receptor agonists.

MetaVia is also developing vanoglipel, a candidate targeting metabolic dysfunction-associated steatohepatitis (MASH), which has shown positive effects on glucose and lipid control in preclinical studies.
