Ultra-Long-Acting Injectable GLP-1 RA Shows Promise for Supporting Weight Management in Individuals With and Without Type 2 Diabetes
New research highlights the potential benefits of berobenatide, an investigational injectable glucagon-like peptide-1 receptor agonist (GLP-1 RA), in improving weight loss among individuals with excess weight or obesity, regardless of type 2 diabetes status. The data were presented during symposium sessions at the 2026 Scientific Sessions of the American Diabetes Association (ADA) in New Orleans.
Obesity and diabetes are associated with an increased risk of serious health conditions such as cardiovascular diseases. As treatment options evolve, researchers are exploring next-generation dosing strategies aimed at enhancing efficacy, simplifying treatment regimens, and lightening the burden of injection load. Berobenatide, which helps regulate appetite and blood glucose, was evaluated with both weekly and monthly dosing options.
The VESPER-3 trial, a phase 2b study, assessed the viability of shifting berobenatide dosing from weekly to monthly while maintaining effective weight loss and tolerability. The results indicated that a monthly dosage of 4.8 mg led to a placebo-adjusted weight loss of up to 12.3% after 28 weeks of treatment. Safety and tolerability were consistent with the GLP-1 RA class.
Dr. John Buse, the lead author of the study, stated, "Berobenatide is a potentially important new option for people living with overweight or obesity, especially because a once-monthly dosing could offer a more manageable alternative to today's more frequent injections." The final endpoint of the VESPER-3 study will take place at 64 weeks and aims to provide long-term data.
Additionally, the VESPER-1 and VESPER-2 trials further demonstrated the positive effects of berobenatide. The VESPER-1 trial indicated that weekly dosing of berobenatide yields effective weight loss and tolerability, while the VESPER-2 trial confirmed a favorable risk/benefit profile for individuals with overweight or obesity, including those with type 2 diabetes.
